What is 5-Amino-1MQ?

5-Amino-1MQ (5-amino-1-methylquinolinium) is a small, positively charged quinolinium compound that selectively inhibits nicotinamide N-methyltransferase (NNMT). It is not a peptide. NNMT is a cytosolic enzyme, abundant in liver and adipose tissue, that transfers a methyl group from S-adenosylmethionine (SAM) to nicotinamide, producing 1-methylnicotinamide and removing nicotinamide from the NAD+ salvage pathway.

This lowers 1-methylnicotinamide production and preserves both substrates: nicotinamide is retained for NAD+ synthesis via NAMPT, and SAM remains available for other methylation reactions.

In adipocyte and animal models the resulting rise in NAD+ and SAM is associated with increased sirtuin-dependent signalling, altered histone methylation and a shift toward oxidative metabolism, with reduced lipid accumulation reported in cultured fat cells.
