What is CJC-1295 No DAC?

CJC-1295 No DAC is a synthetic 29-amino-acid analogue of growth hormone-releasing hormone (GHRH) based on the GRF (1-29) sequence, with four substitutions at positions 2, 8, 15 and 27 (D-Ala, Gln, Ala and Leu). It is more commonly known in the literature as Modified GRF (1-29) or tetrasubstituted GRF (1-29), and it acts as an agonist at the GHRH receptor (GHRHR).

The substitutions were designed to protect the peptide from dipeptidyl peptidase-4 and from oxidation and deamidation, so CJC-1295 No DAC is considerably more stable than sermorelin while retaining a short duration of action. Unlike CJC-1295 with DAC, it has no albumin-binding linker, so it produces a discrete GH pulse rather than sustained elevation.

GenoPept supplies CJC-1295 No DAC in the UK as a lyophilised research peptide in 2mg, 5mg and 10mg vials. Every batch is verified by independent HPLC and LC-MS testing and is supplied strictly for laboratory research.

CJC-1295 No DAC research applications

Pulsatile GH release models

CJC-1295 No DAC is used in pituitary cell and animal models to trigger a single, defined GH pulse, making it well suited to studies of pulse amplitude and timing.

Synergy with ghrelin receptor agonists

The most common research pairing is with ipamorelin or GHRP-2, where the GHRH and ghrelin receptor pathways are studied together for additive GH release.

Peptide stability research

Because each substitution addresses a known degradation route, Modified GRF (1-29) is a teaching example for protease resistance, deamidation and oxidation in peptide design.

GHRH receptor pharmacology

The compound is used as an agonist in GHRHR binding, cAMP and desensitisation assays alongside sermorelin and tesamorelin.

Comparative pharmacokinetics

Research compares the short-acting profile of CJC-1295 No DAC with the multi-day profile of CJC-1295 with DAC to separate the effects of pulsatile and continuous GHRH signalling.

Mechanism of action (preclinical)

CJC-1295 No DAC binds the GHRH receptor on anterior pituitary somatotrophs. This Gs-coupled receptor activates adenylyl cyclase, raising cAMP and activating protein kinase A, which stimulates GH gene transcription and the release of stored GH into circulation.

The D-Ala2 substitution blocks DPP-4 cleavage, Gln8 reduces asparagine deamidation, Ala15 improves receptor affinity and Leu27 prevents methionine oxidation. In preclinical models this gives a half-life of roughly 30 minutes, long enough for a strong GH pulse but short enough that normal somatostatin feedback is preserved.

Reconstitution, handling and storage (laboratory)

CJC-1295 No DAC is supplied as a lyophilised powder that is stable for long-term storage at -20 °C. Unopened vials may be kept at 2-8 °C for short periods, protected from light. Allow a vial to reach room temperature before opening to avoid condensation on the powder.

For reconstitution, add bacteriostatic water or a sterile diluent slowly down the inside wall of the vial, then swirl gently until the powder dissolves. Never shake a peptide vial, because mechanical stress can damage the molecule and reduce measured purity.

After reconstitution, keep the solution at 2-8 °C and use it within the period stated in your laboratory protocol. Full guidance is available in our articles on how to reconstitute research peptides and how to store research peptides.

Quality assurance and Certificate of Analysis

Every batch of CJC-1295 No DAC sold by GenoPept is tested by an independent ISO/IEC 17025 laboratory (Puralytix). HPLC confirms purity of 99% or higher and LC-MS confirms molecular identity, so the compound in the vial matches the label.

Each product page carries a “View Certificate of Analysis” button, and the complete library is published at genopept.co.uk/coa-certificates. Vials are batch-identified with lot number and expiry date, and our guide to what a Certificate of Analysis shows explains how to read the report.

CJC-1295 No DAC frequently asked questions

What is CJC-1295 No DAC used for in research?

CJC-1295 No DAC is used in laboratory research as a stabilised, short-acting GHRH receptor agonist. Typical applications include pituitary and animal models of pulsatile GH release, synergy studies with ghrelin receptor agonists such as ipamorelin, GHRH receptor assays and peptide stability research, where its four substitutions illustrate protease and oxidation resistance. Supplied for in-vitro research only.

How is CJC-1295 No DAC supplied and what purity does GenoPept provide?

GenoPept CJC-1295 No DAC is supplied as a lyophilised powder in sealed, sterile glass vials of 2mg, 5mg or 10mg. Each batch is analysed by an independent ISO/IEC 17025 laboratory, with HPLC confirming purity of 99% or higher and LC-MS confirming identity. The batch Certificate of Analysis can be viewed on the product page or in the COA library.

How should CJC-1295 No DAC be stored?

Keep unopened lyophilised CJC-1295 No DAC at -20 °C for long-term storage, or at 2-8 °C for short periods, protected from light. After reconstitution with bacteriostatic water, store the solution at 2-8 °C and use it within the period stated in your laboratory protocol. Avoid freeze-thaw cycles, add diluent slowly down the vial wall and never shake.

Is CJC-1295 No DAC for human use?

No. CJC-1295 No DAC from GenoPept is a research compound for in-vitro laboratory use only. It is not a medicine, is not for human or veterinary use, and is not for diagnostic or therapeutic purposes. GenoPept does not provide dosing or administration guidance, and all purchasers must be buying for legitimate research use within the UK.

Is CJC-1295 No DAC the same as Modified GRF (1-29)?

Yes. CJC-1295 No DAC and Modified GRF (1-29) are two names for the same tetrasubstituted 29-amino-acid GHRH analogue. The name CJC-1295 originally referred to the DAC-conjugated compound, and the shorter, short-acting version became known commercially as CJC-1295 without DAC. Modified GRF (1-29) or Mod GRF 1-29 is the more precise name used in research contexts.

Research use only. This product is supplied for in-vitro laboratory research only. It is not for human or veterinary use, not a medicine, and not for diagnostic or therapeutic use. See our research use only policy and UK legal guide.
